Abstract


Abstract: Protease is an industrial enzyme with approximately 60% of all enzyme sales in the world. The aim of this study are to isolate and characterize a protease from thermiphilic bacteria from hot springs Pancuran Tujuh Baturraden. Research steps include the isolation of protease producing bacteria, and determine the genus, determine optimum time of protease production, protease isolation, protease purification by ammonium sulfate precipitation and gel filtration. The highest activity protease were characterized, including optimal pH and temperature, and the influence of some metals on the activity.

The results of this study showed that the genus of protease producing bacteria obtained from the hot spring Pancuran Tujuh Baturraden is Bacillus sp. Optimum protease production time approximately 36 hours with 2.116 activity units/ml. The highest protease activity is after precipitation with 15% ammo­ni­um sulfate fraction. Purification by gel filtration using Sephadex G-150 produced the highest activity at fraction of 27. The crude extract protease and of purified protease has an optimum pH of 11, but optimum temperature of 60 and 65 0C.
